The role of haemopoietic colony- stimulating factors in the management of newborn infection

چکیده
Characteristics of neonatal neutrophils Polymorphonuclear neutrophils (PMN) are key components of the innate immune system, provide first line immune defence mechanisms against pathogenic challenge, and are both quantitatively and qualitatively deficient in neonates. Neutrophil deficiencies contribute significantly to the increased susceptibility of neonates to serious and overwhelming bacterial and fungal infections. Although circulating absolute neutrophil counts (ANC) may not differ greatly from those in older children and adults, the release of marrow reserves in response to systemic infection can be markedly impaired as a result of depletion of the neutrophil storage pool, held in reserve within the bone marrow. Severe neonatal sepsis may thus result in neutropenia, often with fatal consequences.

THE ROLE OF ALVEOLAR MACROPHAGES IN THE PRODUCTION OF COLONY –STIMULATING FACTOR BY THE LUNG
The role of alveolar macrophages in the production of granulocyte/ macrophage colony-stimulating factor(s) by the rat lung was investigated. Lavaged lungs, when incubated at proper weight per volume of culture medium, produced the same amount of colony-stimulating factor as unlavaged ones. Both lavaged and unlavaged lungs produced similar types of colony-stimulating factor (s). Prolonged i...

Two years ago we reviewed the rationale for the use of haemopoietic colony stimulating factors (CSFs) in preterm neonates. Two small pilot studies had shown that treatment with either granulocyte colony stimulating factor (G-CSF) or granulocyte-macrophage colony stimulating factor (GM-CSF) could increase the neutrophil count without apparent short term toxicity. 3 Follow up at 2 years of age of...
متن کاملImmunotherapy in patients with systemic mycoses: a promising adjunct.
Evidence from several in vitro and animal model studies suggests a modulatory role of haemopoietic, T(H)1 and T(H)2 cytokines in host defence against fungi, and highlights their potential utility as adjunctive therapy for management of systemic mycoses (SM). However, there are limited clinical data to support the use of cytokines in prevention and treatment of SM. Thus, at present no adjunctive...

The role of growth factors in promoting the survival, proliferation and development of multipotent haemopoietic stem cells and their more developmentally restricted progeny has been unequivocally established using short-term in vitro clonogenic culture systems (Fauser and Messner, 1979; Johnson, 1984; Toogood et al., 1980). In other culture systems, however, haemopoiesis occurs in the absence o...
